At least 1,500 fans have already paid £25 for a ticket to see the band on Sunday at the University of Central Lancashire's Burnley College campus.
The group tweeted to their 330,000 followers that they were supporting Stephen's fundraising campaign, which currently stands at £3.15m.
They said: "All profits from the Lawson gig on 4th May are being donated to the Stephen Sutton appeal.
"See us live at UCLAN Burnley and help support an incredibly worthwhile cause."
The band, who are named after Liverpool-based surgeon Dr David Lawson, who performed life-saving brain surgery on lead singer and guitarist Andy Brown, are set to be the big name band to perform at Burnley College.
Other celebs who have backed 19-year-old Stephen's charity campaign include comic Jason Manford, who raised £10,000 by donating proceeds from a sell-out gig.
Stephen, from Burntwood, Staffordshire, was diagnosed with bowel cancer aged 15, and published a bucket list of dreams he wanted to fulfil before he dies, which included getting a tattoo and raising £1million for charity.
Since his campaign went viral, he has raised more than three times that amount for the Teenage Charity Trust, and has received support from dozens of celebrities, including Robbie Fowler and One Direction's Louis Tomlinson.
