Spurs announced that Townsend will undergo surgery on Thursday and is expected to miss up to 10 weeks.
The 22-year-old came on for a substitute against Stoke on Saturday but left the stadium wearing a protective boot.
After an MRI scan on Monday, further tests on Tuesday confirmed the winger's fate.
Townsend made his England debut earlier this season, scoring on debut against Montenegro in October.

The youngster also played a pivotal role in helping the Three Lions beat Poland to secure World Cup qualification.
But with the tournament now just 43 days away, the injury removes any hope that Townsend had of being included in Roy Hodgson's squad for Brazil.
Townsend was already a long-shot to be included after failing to start a match for Tottenham since the 1-0 defeat to Arsenal in March.
Townsend made 33 appearances for Spurs this season before the injury brought a premature end to his season and his World Cup dreams.
