Townsend came off the bench against the Potters on Saturday to help Tim Sherwood's side to victory.
But the 22-year-old left the Britannia Stadium in a protective boot and underwent an MRI scan yesterday.
Townsend will have further assessments today, but his chances of making the plane to Brazil are now thrown into doubt.

The London-born flyer exploded onto the international stage in October, netting on his England debut against Montenegro before helping Roy Hodgson's men overcome Poland and secure their place in Brazil.
However, Townsend has struggled to match his form at club level this term, with his last Premier League start coming in the 1-0 defeat to Arsenal on March 16.
The Spurs academy graduate has made 33 appearances in all this campaign, having impressed on loan at QPR last season.
